This video demonstrates the spread plate technique, a common microbiological technique for enumeration of aerobic bacteria. This video shows inoculation from a broth culture, but an unknown liquid sample can also be used as your initial inoculum. Our laboratory uses metal spreaders, but glass spreaders can also be used in the same manner.
